The Dodgeball star, 48, was pulled over in Manhattan Beach, California on 10 June by police officers, but refused repeatedly to get out of his car.
Police said he failed a field sobriety test, captured on a body-worn camera.
A blood test showed he was over the legal limit, said authorities.
Los Angeles County prosecutors have now charged him with driving under the influence of alcohol, driving with a blood alcohol level of .08 or higher, and refusing to comply with police.
Vaughn, whose films include Wedding Crashers, The Break-Up and Anchorman, has not entered a plea and is due to appear in court to be arraigned on the charges on Monday.
